Not necessary to plug off. Regardless, you will have to bleed a new caliper so, plugging is not needed.
It will help to keep the fluid from running out all over everything and save 'some' bleeding, depending on how fast you are!
ask your auto parts store they can help you.
Hose clamping tool on the rubber flex hose,..
