Misfire cylinder no. 5, I have tried everything I can think of, spark plugs, coil packs, plug wires, timing belt kit with new tensioner, no vacuum leaks.

Well now how about the fuel injector???? ..... Injector can cause misfiring just like ignition!!
Hello, if you have verified that spark and fuel are present, the next thing to do is to check for engine compression. Have a compression test performed and a cylinder leak down test. This will tell you if that number 5 cylinder is weaker than the rest mechanically internally, and can also tell you if there is a problem in the valve train or the base engine before you go tearing it apart to research it further. Yes, I did check the injectors and they checked out ok, I did forget to mention however that it was misfiring on cylinder 1 and 5, that is when I bought coil packs thinking that was the problem and now it is just misfiring on 5.
Make SURE you put the new coil on the proper cylinder!!! .. Mighty easy to get it wrong... Just saying. Cylinder layout on firewall side - front to back is > 1---3---5
Yes I know your right about that, it is easy to get it wrong, I bought 3 new coil packs to make sure, yes it is 1, 3 ,5 other side 2,4 and 6, 1 goes to 4, 3 goes to 6, and 5 goes to 2.
We never know ones skill level .. Can only make suggestions, it's all good! Make sure misfire is actually on cylinder # 5 ... It may be on another cylinder.
I appreciate any advice and input given, I did not mean to sound like a know it all, sorry, I am only going off of what the scan tool is telling me for being cylinder 5, but you never know the ecm could be saying that and it could be a different one, I just know it is frustrating.
Well now a scanner is definitely a big help! .. IF scanners live misfire data screen is showing # 5 misfire while engine is on then that is most likely correct .. Go back to the basics, fuel - fire & compression. .. 'LONG' screwdriver touching injector with ear on handle, can you hear injector pulsing/clicking ?... Use screwdriver 'in a spearing motion' to lightly tap injector to see if engine smooths out... You got to be right on top of the problem, you'll find it.
I am going to try that, I really appreciate all of the help, I have read a lot of articles on this issue and I cannot understand why a lot of them want to point towards bent valve, because this car is in like brand new condition engine wise, its never been in a wreck, it has never had any engine work except for what I am having to do now, and oil levels and fluids are checked daily and they are always right on full line, so I do not see how a valve would get bent. I also have another question, I have a 95 mustang 3.8 v6, it makes a whistling sound when just barley pressing on gas pedal, like it is sucking in air, I have tried spraying carb cleaner everywhere and vacuum gauge is in green normal engine range?
