The Prius has a brand new battery and after stopping and shutting down the engine it will not start again without being Jumped. All electrical systems are off what drains a battery so quickly. What shorts the system so quickly and completely? After jumping the battery the car runs fine. The time between shutoff and dead in less the 30 seconds.
How long has your 2010 Toyota Prius had this problem?
