Hello, I’m Pete from the Car Genius team at RepairPal. I’m sorry you’re dealing with this concern on your Lexus GS350. The "S" or Sports mode will stop engaging anytime the computer systems detect an issue with one of the transmission components whether electrical or mechanical, a shifter concern, or in some cases when certain DTC's (fault codes) are stored in the computer systems. I recommend having the fault codes checked using a high end scan tool that most shops use to help identify any related codes to this concern. The scan tool can also be used to monitor transmission signals and shifter signals to help pinpoint the cause. Once the cause has been identified it can then be repaired properly based on which component is at fault.
