Sounds like your having problems with the shift lock system. There are a few reasons this may be happening, so it's best to have it looked at by an experienced MB tech.

I get the same problem with my 2003 Mercedes S430. When I start my car and try to shift into reverse, it locks up and I can't move the shifter. Then when I turn off the car to try again, it works. It has been a big pain as I have to start the car, try to shift, turn off the car and start again to shift into reverse. Appreciate any new info to this problem.

Just my luck I made this $1200+ repair last month @ MB of LB -- parts required were the Shifter, Pin, and Lock. Removel and replacement are made inside the car. It did not look hard for a DIY, I was going to do it myself; however, I was not sure the shifter replacement was the answer. And I would not have not been prepared/aware of the lock and the pin replace. Hope you can diy and save - good luck
